We currently provide four types of services:

  1.  DNA preparation.

  2.  DNA injection to produce transgenic flies: P-element, site-specific and MiMIC.

  3.  Selection of transgenic flies following DNA injection.

  4.  Balancing of transgenic insertions following selection

Each of these services is discussed in detail below.

1. DNA preparation.(return)

We offer DNA preparation services for any type of construct. The pricing is as follows:

  1. For DNAs less than 20 kb, the cost is only $30 per DNA.

  2. For DNAs between 20 and 40 kb, the cost is $40 per DNA. We strongly urge that you use our DNA preparation services for DNAs of this size as this is a critical factor in determining the rate of success of transgenesis and we have perfected preparation of large DNAs for injection.

  3. For DNAs greater than 40 kb, we require that we prepare the DNA for you and the cost is included in the total cost of injection (see our Pricing page for full details).

2. DNA injection to produce transgenic flies.(return)

We currently provide two types of injections:

  1. Standard P-element mediated transgenesis

  2. PhiC31 integrase-mediated site specific transgenesis. We can inject into any of the following docking sites at no additional charge:     VK37: (2L) 22A3     P1:     (2R) 55C2     VK1:   (2R) 59D3     VK31: (3L) 62E1     P2:     (3L) 68A4     VK20: (3R) 99F8

In addition, we can inject into any other docking stock you choose, but there is a $50 setup fee for each stock, you will usually provide us with the flies, and there will be a 2-4 week delay while we expand the stock for embryo collection. A full list of docking stocks that can be used is available at the Bloomington Stock Center website: http://flystocks.bio.indiana.edu/Browse/phiC31.htm. For the list of references for the docking stocks, go to the resource page.

3. Selection of transgenic flies following DNA injection.(return)

For injections of DNAs less than 40 kb, we can set your G0 crosses and screen for white-plus transgenics, saving you the time and bother of doing it yourself. Our costs are so low that it is probably less expensive to outsource this step to us instead of paying someone in your lab to do it. In addition, since we are selecting the transgenics and we guarantee our work for constructs less than 40 kb in length (including vector sequences), you know that once you send your DNA, your transgenics will be on the way.

4. Balancing of transgenic insertions following selection.(return)

For PhiC31 injections, we will balance two independent transgenic insertions over either FM7, CyO, or TM3/TM6B, depending upon which docking stock you have chosen. For insertions in VK31 (3L)62E1, we will use TM6B since TM3 does not balance the distal half of 3L well. For all other third chromosome docking stocks we will use TM3. This service costs $30.

For P-element insertions, we will map and balance at least three independent insertions. Since mapping adds considerable time and effort, this service costs $100.
